German and Brazilian researchers have announced that they have found a new way to cultivate Zika virus in the laboratory, which will help virus research and vaccine preparation.

Since May 2015, Zika virus has spread in Brazil and other countries in the United States, and vaccine development is imminent. The large-scale cultivation of viruses in laboratories is an important prerequisite for mass production of vaccines.

The virus lacks a complete enzyme system and organelles, and must be parasitized in living cells to proliferate. At present, African green monkey kidney cells and Aedes albopictus larvae cells are commonly used in the medical community as cell hosts for culturing viruses. Experts from the Institute of Complex Technology System Dynamics of the Max Planck Society of Germany chose the BHK-21 cell line (young hamster kidney cells) and allowed them to grow in liquid medium.

The study found that the BHK-21 cell line is very suitable for Zika virus proliferation, but the Zika virus isolates from different regions have different virus yields after infection, and not all strains can produce high yield.

In the existing culture environment, the flavivirus to which the Zika virus belongs can only form about 10 infectious virus particles in each host cell. In contrast, influenza viruses can form hundreds of infectious viral particles in each host cell. Therefore, if you want to produce Zika vaccine efficiently, you should either look for a higher-yielding cell line or work hard to increase cell concentration.

Nowadays, researchers have increased the cell concentration to six times that of traditional culture methods, and created an environment suitable for virus proliferation by adjusting the temperature, oxygen content and pH of the bioreactor. After nearly two weeks of incubation, the researchers harvested 40 million infectious viruses per ml of culture fluid, enough for a large number of virological studies.

The researchers said that although people have already successfully used the BHK-21 cell line to produce veterinary vaccines, whether they can be used to produce human vaccines still needs further study. However, with the new method of "production" virus, Zika virus has been able to conduct in-depth research on infectious diseases and virology.
