Free circulating DNA (cfDNA) is present in the blood and contains double-stranded DNA fragments, most of which are short-chain, and usually at low concentrations. In healthy humans, plasma cfDNA is thought to be derived from normal hematopoietic cells. The circulating half-life of cfDNA is very short, suggesting that it requires sustained release during apoptosis. In certain physiological conditions or diseases, a considerable part of cfDNA is different from when it is healthy. Based on this result, cfDNA has been used for non-invasive diagnosis in recent years. Among pregnant women, about 10%-15% of cfDNA comes from the placental trophoblast. Now cfDNA is used to screen fetal defects in high-risk pregnant women. This is the basis of the current hot non-invasive Down's screening! In cancer patients, detection of tumors by quantitative mutations in cfDNA has been recognized by more and more people. In terms of transplantation, transplant rejection is also associated with cfDNA levels in donor transplanted organs.

Circulating tumor DNA (ctDNA) is a type of cfDNA derived from tumor cells and belongs to a type of cfDNA. Both are inclusive and ctDNA is only a small part of cfDNA. ctDNA is derived from somatic mutations in tumor cells, unlike genetic mutations that are present in every cell in the body. Often when it comes to circulating tumor DNA, people tend to think of it as free DNA in the peripheral blood of cancer patients.
