1. Ultra-clean workbench : At present, most cell laboratories use ultra-clean workbench to achieve aseptic operation. It has simple operation, convenient installation, small space and good purification effect. There are two main types of ultra-clean work benches: side flow (vertical) and outflow (horizontal laminar flow). The working principle is generally to filter the indoor air through the coarse filter, press it into the static pressure box by the centrifugal fan, and then finely filter through the high-efficiency air filter, so that the clean air flow sent through the sterile zone with a certain uniform cross-section wind speed. Thereby forming a dust-free and sterile high-clean working environment.

(1) Side-flow workbench: airflow after air purification flows from the left or right side through the work surface to the opposite side, and also flows from the top to the bottom or from the bottom to the opposite side, forming an airflow barrier to keep the working area sterile, and the workbench The structure is closed;

(2) Outflow type workbench: The purified air flows toward the operator, so the external airflow is not mixed into the operation, and the workbench structure is open, but the experimental operation of the harmful substances is unfavorable to the operator. The ultra-clean workbench should be inspected regularly by the relevant departments to check the cleanliness. The cleanliness of the ultra-clean workbench that meets the requirements should reach 100. The dust particles with a particle size of ≤5μm should not exceed 3.5/L. The air flow rate should be controlled at 0.75-1.0m3/s; the average number of bacterial colonies is <1, and the filter should be replaced if necessary according to the sterility condition.

2. Microscope : The inverted microscope is an essential equipment for routine work in the cell culture laboratory. It is mainly used to understand the growth of cells and observe the presence or absence of pollution. If funds permit, it is recommended to use a high-quality phase contrast microscope, a dissecting microscope, a fluorescence microscope, a video system or a time-lapse movie camera equipped with a camera system to record and record cell growth at any time.

3. Incubator : The cells cultured in vitro need to survive at a constant temperature like the cells in the body. Generally, the optimum growth temperature is 37 °C, and the temperature difference should not exceed ±0.5 °C. When the temperature rises by 2 °C, it is not conducive to cell survival. When the temperature reaches 40 °C, the cells will die very quickly. Therefore, a thermostatic incubator and a CO2 incubator with precise temperature control are the best choices.

(1) Constant temperature incubator: A water-blocking or transistor-type self-regulating incubator should be selected. This type of incubator has high sensitivity and stable temperature control. The general constant temperature incubator is cheaper, and the disadvantage is that it is only suitable for closed culture.

(2) CO2 incubator: Most cell culture chambers are currently widely used. The advantage of the CO2 incubator is that it can provide a certain amount of CO2 (usually 5%) required for cell culture, and it is easy to stabilize the pH of the culture solution, and is suitable for open or semi-open culture. When using the culture bottle, in order to keep the inside of the culture bottle ventilated, the cap can be slightly loosened. To avoid contamination of the cells, the air in the incubator must be kept clean when using this culture method. Or alcohol disinfection, while the incubator should be placed in a tank containing sterile distilled water to prevent evaporation of the culture solution, keeping the relative humidity inside the box at 100%.

4. Oven (drying box) : Some instruments and utensils used for cell culture must be dried before use, and glassware should be dry and heat sterilized. In dry heat sterilization, the temperature in the oven is generally above 160 ° C, usually using a blown oven. The advantage is that the temperature is uniform and the effect is good, and the disadvantage is that the heating process is slow. When the temperature is raised, the blast may not be heated first, but the blasting and the temperature increase shall be started at the same time. When the temperature is raised to 100 ° C, the blast is stopped. Care should be taken to avoid burning paper or cotton in the wrapper, which can affect cell growth. After disinfection, the door cannot be opened immediately to prevent the glassware from rupturing due to quenching. The door should be opened after the temperature naturally drops below 100 °C.

5. Water purification device : Cell culture requires high quality of water, and cell culture, water for preparation of cell culture-related liquids, and water for washing cell culture vessels must be subjected to strict purification treatment in advance. At present, there are a variety of purification methods combined, which can make ordinary water purified into pure water and ultrapure water. The pure water device is very flexible and convenient. It can be wall-mounted, bench-top, can be equipped with a water storage tank, or can be directly used with a liquid separation gun. The bactericidal function can also be selected according to various experimental water requirements, and DNase, RNase, protease, etc. can be effectively removed, and an ultrafiltration type pure water device capable of effectively removing heat source and endotoxin can be effectively removed.

6. Refrigerator : The necessary equipment for cell culture laboratory should be dedicated. It should not store substances that are harmful to cells, such as volatile, easy to burn, and should be kept clean. Generally, it includes a general refrigerator, a low temperature refrigerator, and an ultra low temperature refrigerator. (1) Ordinary refrigerator: Store culture materials such as culture solution, physiological saline, Hanks liquid reagent, etc., and store tissue samples for a short period of time. (2) -20 ° C low temperature refrigerator, ultra low temperature refrigerator: used to store preparations that need to be frozen to maintain biological activity and need to be stored for a long period of time, such as enzymes, serum and the like.

7. Cell Freezer Storage : Liquid nitrogen containers commonly used in storage are classified into different types and specifications according to the needs of use. There are three factors to consider when selecting a liquid nitrogen container: volume, ease of handling, and liquid nitrogen volatilization. The size of the liquid nitrogen container is generally 25-500L, and can store about 12.5 ampoules of 250-15000. The lowest temperature of liquid nitrogen can reach -196 °C, and should be used to avoid frostbite. Since liquid nitrogen is volatile, it is necessary to observe the residual liquid nitrogen amount and replenish it in time to avoid damage or death of the cells caused by insufficient liquid nitrogen. There are many smart cell cryopreservation options available, and a liquid nitrogen storage device with an electronic controller can be configured to automate the cryopreservation; liquid nitrogen levels and sample temperatures can be monitored to ensure that the sample temperature is always at the set temperature point; It can be configured with an alarm system to set alarms for abnormal liquid nitrogen, temperature, battery, voltage, power supply, etc. It also has a hot gas bypass system to prevent warm air above -130 °C from entering the liquid nitrogen tank, thus more effectively Protect the sample from heat inside the container. In addition, the liquid nitrogen supply tank can be selected to replenish the storage tank with liquid nitrogen through the connecting pipe to ensure the safety of the sample.

8. Centrifuge : In cell culture, a cell suspension is usually prepared using a centrifuge, cell density is adjusted, and cells are washed and collected. A 4000 rpm benchtop centrifuge is generally conventionally configured; if cell sedimentation is to be performed, 80-100 G centrifugal force is required because the centrifugal force excessively damages the cells. According to different requirements, there are more functions such as large capacity, adjustable temperature centrifuge, high speed centrifuge and cryogenic refrigerated centrifuge.

9. Balance : Commonly used are precision balances and analytical balances. The accuracy of the analytical balance is generally 0.1 mg, 0.01 mg and 0.001 mg. Generally, the appropriate balance should be selected according to the sampling quantity and precision requirements: the sampling volume should be more than 100mg, the balance should be selected with an accuracy of 0.1mg; the sampling amount should be 100-10mg, and the balance with the precision of 0.01mg should be selected; the sampling amount should be 0.001mg. Balance. The balance needs to be calibrated regularly, and a balance with automatic calibration can be selected for easy maintenance.
