Cactus Tissue Culture Rapid Propagation Technology

<

The edible cactus is a perennial thick-fleshed, multi-plasma plant of the genus Cactaceae and Cactus. It is native to the Americas and is rich in Mexico. It is a special variety of high-quality vegetables and contains many vitamins and traces of calcium, phosphorus, and iron needed for human health. Elements, and contains amino acids, cellulose, succinate and other nutrients, with ventilation and blood circulation, detoxification of heat. The Chongqing Institute of Agricultural Sciences used the excellent edible cactus variety "Mibanga" as a special health vegetable for introduction and domestication cultivation. The results showed that: "Mibongta" has a well-developed water storage structure and good resistance to stress in Chongqing. The city grows well under special natural and ecological conditions and has a good development prospect. The edible cactus is usually propagated by the ramets method, and the reproduction rate is low, and the shoots are easily rotted and the survival rate of the plants is low. In order to speed up the popularization and application process of edible cactus, we have carried out research on the tissue culture and rapid propagation of edible cactus and achieved good progress. Scientists and technicians have systematically studied the multi-factors affecting the rapid propagation of cactus in tissue culture, and established the technical regulations for rapid propagation of seedlings: 1 Adventitious buds are induced to grow. Take healthy natural month-old seedlings, disinfect and sterilize, cut into small pieces of 0.5 square centimeters, and inoculate them on MS medium supplemented with high-concentration hormones under aseptic conditions for induced growth and culture. It is 40--50 days. 2 subculture and proliferation. About 1 cm of adventitious shoots were excised and inoculated on proliferation medium with reduced hormone concentration. Subculture was performed once every 30-35 days, and the propagation coefficient was 6-8. 3 rooting induction culture. More than 1 cm of shoots were taken and inoculated on 1/2MS rooting medium. Rooting was started 7 days later. After 30 days of culture, the shoot height was above 4 cm and the rooting induction rate was 98%. 4 hardening seedling transplanting. After 4 centimeters of rooted seedlings were planted for 3 days, seedlings were transplanted in a meadow soil with a sediment ratio of 1:1, and maintained at a relative humidity of about 80% for 7 days. The survival rate of the seedlings was 96%. Through this technical regulation, the large-scale production of edible cactus seedlings can be carried out in a relatively short period of time. At the same time, the technical basis for the rapid promotion and application of the special health vegetable “Mibongta” has been established. China Agricultural Network Editor
